*Spermatids (Immature Sperm Cells) can be differentiated from polymorphonuclear white blood cells (WBC) by using the following criteria: WBC contain connecting nuclear bridges and have a granular cytoplasm; Immature Sperm Cells are more spherical in shape.
Summary
STAT III Andrology Stain is a convenient manual stain set for staining sperm. The staining characteristics are similar to the traditional Wright's and Wright's Giemsa stain and meet the Kruger Strict Criteria for Sperm Morphology.
STAT III Andrology Stain offers the flexibility of a three to seven dip staining sequence in both stain solutions. Excellently stained slides exhibiting high contrast and definitions are produced in less than thirty seconds.
Return to Top Storage Conditions
Store at controlled room temperature (15-30°). STAT III. Containers should be re-closed between use. Do not use after expiration date on container.
Specimen Collection and Preparation
Smears prepared from fresh or thawed semen should be air-dried for five to fifteen minutes.
Procedure
- Prepare slides as instructed above.
- Dip slide three to seven times (one second each dip) in Fixative. Touch edge of slide to inside of Fixative chamber to allow excess solution to drain and blot edge of slide on an absorbent paper towel.
- Dip slide three to seven times (one second each dip) in Solution I (eosin solution). Touch edge of slide to inside of Solution I chamber to allow excess solution to drain and blot edge of slide on an absorbent paper towel.
- Dip slide three to seven times (one second each dip) in Solution II (azure solution). Touch edge of slide to inside of Solution II chamber to allow excess solution to drain and blot edge of slide on an absorbent paper towel.
- Rinse slide with deionized water.
- Allow to dry and examine under oil immersion.

- An important advantage of the STAT III Andrology Stain is the flexibility to produce desirable results for varied applications. Staining intensity may be varied by increasing or decreasing the number of dips in eosin and azure stain solutions.
- Eosinophilic staining (red tones) may be intensified by increasing dips in Solution I (eosin solution).
- Basophilic staining (blue tones) may be intensified by increasing dips in Solution II (azure solutions).
- Three to seven dips in each of Solutions I and II will provide desirable results for most staining applications.

- Any variation of the procedure other than described above may adversely affect results.
